Hypothetically, by increasing the cell uniformity in the scaffold, the restorative properties of an engineered-ligament is also increased, making it a more viable option for ligament reconstruction in the future. The purpose of this study is to find an optimized cell seeding protocol, which will include the design and manufacture of a ligament-specific seeding and culture vessel. The vessel will allow the silk scaffolds to remain aligned during cell seeding and culture and promote uniformity of cell distribution. Furthermore, this study will focus on optimizing two types of active distribution centrifugal and vacuum pressure cell seeding.
